Chrysler's Eastwood Super Bowl Ad Praised by Obama Democrats (ContributorNetwork)

COMMENTARY | Chrysler's two-minute Super Bowl halftime ad with Clint Eastwood is being condemned as being inappropriately pro-Barack Obama's re-election by Karl Rove and other conservative pundits.

Chrysler CEO Sergio Marchionne denies any political intent and notes Eastwood's gravelly praise of a Detroit "comeback" does not mention President Obama.

Still, I won't join liberals who assert Rove is merely engaged in his usual whining and spinning. This time the operative once known as Bush's Brain has a point, although he perhaps protests too much in pleading to his friends at Fox, "(I)t is a sign of what happens when you have Chicago-style politics, and the president of the United States and his political minions are, in essence, using our tax dollars to buy corporate advertising."

C'mon, Rove! Deep dish pizzas and hot dogs with pickles may be "Chicago-style," but that's such an old-hat charge against Obama, you'd think he was the old Mayor Daley or someone. This ad was cast in Detroit, as was one for last year's Super Bowl featuring, in contrast to Eastwood, the hip-hopper Eminem. Also keep in mind that Eastwood is (or at least used to be) a libertarian Republican who filmed "Gran Torino" in Detroit.

Nonetheless, was there a pro-Obama slant to the Super Bowl halftime ad, intended or not? Well, three Obama spokesmen -- David Axelrod, Dan Pfeiffer and Bill Burton (now head of his super PAC) -- quickly twittered how great they felt about the ad. This shows why they've been lousy spokesmen who generally haven't served Obama well. If they had any smarts, they would have kept their mouths shut rather than playing into Rove's hands.

Chrysler's ad team, meanwhile, would have done better to run a spot that included a thank you to the American taxpayers, a note of the number of job saved and a vow to continue repayments on the bailout, which despite proclamations by Chrysler (and General Motors) is nowhere near being fully repaid.

Paris airports downplay early impact of strike

Air France and other airlines have so far limited the damage on Monday from a strike by aviation industry workers by canceling or rescheduling at least 100 flights in advance, Paris aviation officials said.

The strike led by labor unions representing pilots, cabin and ground crews and others didn't draw a large walkout, according to early signs, the Paris airports authority ADP said.

As many as 200 flag-waving protesters marched from terminal to terminal at Paris' Charles de Gaulle airport ? under watch from nearly as many riot police.

French flag carrier Air France said about one in five scheduled flights, or slightly over 100, had been cancelled shortly after midday. Most involved medium-haul flights to European destinations; only 4 were long-haul flights.

Unions called the strike through to Thursday to protest a bill in Parliament that would require air transport workers to give 48 hours notice before striking.

Some airlines were caught off-guard by workers who didn't pre-announce that they'd be on strike: ADP said low-cost carrier easyJet was forced into canceling five flights Monday after employees didn't show.

British Cabinet Warns of Possible Internet Outages During Summer ...

The British Cabinet Office has warned businesses in England that the country may not be able to fully accommodate a surge in Internet use during the 2012 Olympics in London this summer.

Businesses have been told that Internet providers may be forced to ration access to the web during certain times of day and service may fall out altogether at other times.

?A lot of businesses have still not prepared for the enormous risks presented by the London Games,? Kathryn Hurt, an executive at a workspace leasing company, told England?s Guardian newspaper. ?There?s been a lot of discussion about traffic hotspots, but very little about potential internet traffic problems. The risk is that home workers are unable to work effectively due to over-capacity.?

A government document called Preparing Your Business for the Games raises the possibility ?that internet services may be slower during the Games or, in very severe cases, there may be dropouts due to an increased number of people accessing the internet.?

Streaming video has become an increasingly popular way for people to consume content, and the British government encourages businesses to steer clear of data-heavy web usage. ?Consider alternative means of communication (for example, video-streaming may greatly reduce your internal network?s capacity),? the report reads.

The Cabinet Office also encourages businesses to allow employees to work from home and in staggered shifts to lessen the bandwidth burden on high-usage areas.

It?s hard to imagine the Internet simply failing on a large scale like the government report warns of, but the sheer numbers expected for the London Olympics make such a scenario a bit more understandable. Up to 850,000 athletes, spectators, officials, media members and organizers are expected to be travelling to and from Olympic facilities each day, according to the Guardian.

Scientists use old theory to discover new targets in fight against breast cancer

ScienceDaily (Feb. 7, 2012) ? Reviving a theory first proposed in the late 1800s that the development of organs in the normal embryo and the development of cancers are related, scientists at the Salk Institute for Biological Studies have studied organ development in mice to unravel how breast cancers, and perhaps other cancers, develop in people. Their findings provide new ways to predict and personalize the diagnosis and treatment of cancer.

In a paper published February 3 in Cell Stem Cell, the scientists report striking similarities between genetic signatures found in certain types of human breast cancer and those of stem cells in breast tissue in mouse embryos. These findings suggest that cancer cells subvert key genetic programs that guide immature cells to build organs during normal growth.

"Stem cells in a healthy developing embryo have a GPS system to alert them about their position in the organ," says Geoffrey Wahl, a professor in Salk's Gene Expression Laboratory, who led the research. "The system depends on internal instructions and external signals from the environment to tell the stem cell what to do and where to go in the body. It stimulates the stem cells to grow and form more stem cells, or to change into different cells that form complex organs, such as the breast. Our findings tell us that this GPS system is broken during cancer development, and that may explain why we detect stem-like cells in breast cancers."

The relationship between cancer and embryonic tissues was first proposed in the 1870s by Francesco Durante and Julius Cohnheim, who thought that cancers originated from cells in adults that persist in an immature, embryonic-like state. More recently, scientists including Benjamin Spike, a co-first author on the current work and post-doctoral fellow in the Wahl lab, have discovered that tumors often contain cells with stem cell characteristics revealed by their genetic signatures.

As a result, many scientists and physicians are pursuing ways to destroy stem-like cells in cancer, since such cells may make cancer more resistant to treatment and may lead to cancer recurrence. The Salk scientists are now characterizing the stem-like cells in certain forms of breast cancer to arrest their growth.

Studying the genetic activity of organ-specific stem cells is very difficult because the cells are very rare, and it is hard to separate them from other cells in the organ. But, by focusing on tissue obtained from mouse embryos, the Salk researchers were able for the first time to identify and isolate a sufficiently large number of fetal breast stem cells to begin to understand how their GPS works.

The Salk scientists first made the surprising finding that these fetal breast stem cells were not fully functional until just prior to birth. This observation suggested that a very special landscape is needed for a cell to become a stem cell. The breast stem cells at this late embryonic stage were sufficiently abundant to simplify their isolation. This enabled their genetic signature to be determined, and then compared to that of the stem-like cells in breast cancers.

The signatures of the breast stem cells in the fetus were stunningly similar to the stem-like cells found in aggressive breast cancers, including a significant fraction of a virulent cancer subtype known as "triple-negative." This is important as this type of breast cancer has until now lacked the molecular targets useful for designing personalized therapeutic strategies.

"The cells that fuel the development of tumors in the adult are unlikely to 'invent' entirely new patterns of gene expression," says Benjamin Spike. "Instead, some cancer cells seem to reactivate and corrupt programs that govern fetal tissue stem cell function, including programs from their neighboring cells that constitute the surrounding fetal stem cell landscape, or microenvironment."

The discovery of the shared genetic signatures provides a new avenue for scientists to explore the links between development and cancer. By uncovering new biological markers, the scientists hope to develop tests that individualize treatment by showing how the GPS system of a tumor operates. This should help doctors to determine which patients may benefit from treatment, and the correct types of treatment to administer.

Doctors are already using drugs, such as Herceptin, that specifically target malfunctioning genetic pathways in tumors, but no such therapies are currently available for certain aggressive forms of the disease, such as the triple negative subtype.

Although triple negative cancer cells lack the three critical genetic markers that are currently used to guide breast cancer treatment, the scientists' analysis suggests a strong reliance on signaling through pathways similar to those that affect fetal breast stem cell growth.

They found that the fetal breast stem cells are sensitive to a class of targeted therapies that already exists, so these therapies might also work in triple negative breast cancers. Laboratory studies and clinical trials are currently underway to test this possibility.

"Substantial effort is being expended to personalize cancer treatment by gaining a better understanding of the genetics of an individual patient's cancer," Wahl says. "Our findings offer a way to discover new targets and new drugs for humans by studying the primitive stem cells in a mouse."

In addition to Spike, Dannielle Engle and Jennifer Lin, both postdoctoral researchers in Wahl's laboratory, were also co-first authors on the paper.

To explore the most intricate structures of the brain in order to decipher how it functions ? Stefan Hell's team of researchers at the Max Planck Institute for Biophysical Chemistry in G?ttingen has made a significant step closer to this goal. Using the STED microscopy developed by Hell, the scientists have, for the first time, managed to record detailed live images inside the brain of a living mouse. Captured in the previously impossible resolution of less than 70 nanometers, these images have made the minute structures visible which allow nerve cells to communicate with each other. This application of STED microscopy opens up numerous new possibilities for neuroscientists to decode fundamental processes in the brain.

Every day a huge quantity of information travels not only over our information superhighways; our brain must process an enormous amount of data as well. In order to do this, each of the approximately hundred billion nerve cells establishes contact with thousands of neighboring nerve cells. The entire data exchange takes place via contact sites ? the synapses. Only if the nerve cells communicate via such contact sites at the right time and at the right place can the brain master its complex tasks: We play a difficult piece of piano, learn to juggle, or remember the names of people we have not seen for years.

We can learn most about these important contact sites in the brain by observing them at work. When and where do new synapses form and why do they disappear elsewhere? This is not easy to determine, since details in living nerve cells can only be observed with optical microscopes. Due to the diffraction of light, however, structures located closer together than 200 nanometers (200 millionths of a millimeter) appear as a single blurred spot. The STED microscopy developed by Stefan Hell and his team at the Max Planck Institute for Biophysical Chemistry is a groundbreaking method devised to surpass this resolution limit. They use a simple trick: Closely-positioned elements are kept dark under a special laser beam so that they emit fluorescence sequentially one after the other, rather than simultaneously, and can therefore be distinguished. Using this technique, Hell's team has been able to increase the resolution by approximately tenfold compared to conventional optical microscopes.

STED microscopy has already found wide application in fields ranging from materials research to cell biology. Under this microscope, cell cultures and histological preparations have offered unique insights into the cellular nanocosmos. The first real-time video clips from the interior of a nerve cell have demonstrated how tiny transmitter vesicles migrate within the long nerve cell endings.

A vision becomes reality

What was only an ambitious vision a year ago has now become reality: to also study higher living organisms at this sharp resolution in the nanometer range. By looking directly into the brains of living mice using a STED microscope, Hell and his team were the first ones to image nerve cells in the upper brain layer of the rodent with resolution far beyond the diffraction limit.

"With our STED microscope we can clearly see the very fine dendritic structures of nerve cells at which the synapses are located in the brain of a living mouse. At a resolution of 70 nanometers, we easily recognize these so-called dendritic spines with their mushroom- or button-shaped heads," explains Hell. They are the clearest images of these fundamental contact sites in the brain to date. "To make these visible, we take genetically modified mice that produce large quantities of a yellow fluorescing protein in their nerve cells. This protein migrates into all the branches of the nerve cell, even into smallest, finest structures," adds Katrin Willig, a postdoctoral researcher in Hell's department. The genetically modified mice for these experiments originated from the group of Frank Kirchhoff at the G?ttingen Max Planck Institute for Experimental Medicine. Images of the nerve cells taken seven to eight minutes apart revealed something surprising: The dendritic spine heads move and change their shape. "In the future, these super-sharp live images could even show how certain proteins are distributed at the contact points," adds Hell. With such increasingly detailed images of structures in the brain, Hell's team hopes to shed light onto the composition and function of the synapses on the molecular level.

Such insights could also help to better understand illnesses that are caused by synapse malfunction. Among these so-called synaptopathies are, for example, autism and epilepsy. As Hell explains, "Through STED microscopy and its application in living organisms, we should now be able to gain optical access of such illnesses on the molecular scale for the first time." As one of the two representatives of the G?ttingen Research Center Molecular Physiology of the Brain funded by the German Research Foundation, he is committed to collaboration in his further research. Together with neurobiologists and neurologists, he and his team plan to transfer the progress made in imaging technology into fundamental knowledge about the functioning of our brains.

WASHINGTON ? Mitt Romney's remark that he's not worried about the very poor, the latest gaffe in a campaign rich with blunders, joins a long list of wait-let-me-explain episodes in presidential election history.

It's been a banner year for campaign misfires: Rick Perry had his "oops" moment when he forgot one of the three government departments he wanted to eliminate. Herman Cain only made things worse after he fumbled a question about Libya when he explained that he had "all this stuff twirling around in my head." Michelle Bachmann launched her campaign with a cringe-worthy misfire, declaring that both she and actor John Wayne had lived in Waterloo, Iowa, when it was actually serial killer John Wayne Gacy Jr. who'd lived there.

Will any of those sour notes still be ringing in the ears beyond November's ballots and confetti?

There's stiff competition in the pantheon of campaign misfires: Think of Howard Dean's primal scream in Iowa during the 2004 primary. Vice President Al Gore's overwrought sighs when debating George W. Bush in 2000. Vice President Dan Quayle's botched spelling of potato in 1992. And, way back at the dawn of televised presidential debates, Richard Nixon's profuse sweating on stage with cool-as-a-cucumber rival John Kennedy in 1960.

Some others with proven staying power:

THE OTHER ROMNEY. Mitt Romney knows only too well how devastating a single gaffe can be. Forty-five years ago, his father, George Romney, ended his presidential campaign after negative fallout from his answer to a question about why he he'd once supported the Vietnam War. In a 1967 TV interview, Romney referred back to his 1965 visit to the country and stated, "When I came back from Viet Nam, I'd just had the greatest brainwashing that anybody can get." He said he'd since done a lot more study of the matter and no longer believed the war was necessary.

Romney's poll numbers sank amid a swirl of ridicule and questions about whether he was naive. "Can't you just see him coming back from a conference with (Soviet official Alexei) Kosygin yelling that he had been brainwashed by a Russian?" Democratic Party Chairman John Bailey asked. Romney's wife, Lenore, allowed that her husband's words were "extremely unfortunate" and insisted that he was too strong a man to be brainwashed. But the damage had been done.

DEBATE DOMINATION. President Gerald Ford didn't dominate when he falsely declared in a 1976 debate that "there is no Soviet domination of Eastern Europe," including Poland. Time magazine called it "the blooper heard round the world." Democrat Jimmy Carter, Ford's rival, said the president had "disgraced our country." Ford only made things worse by refusing for days to retract the statement and offering clarifications that didn't really clarify things.

At one bizarre campaign appearance, the president spoke to reporters on a press bus via walkie-talkie and referred to himself in the third person, saying: "President Ford does not believe that the Polish people over the long run ? whether they are in Poland or whether they are Polish-Americans here ? will ever condone domination by a foreign force." The incident is recounted in Alan Schroeder's book about presidential debates, "Forty Years of High-Risk TV." Ford eventually apologized and said he recognized that the Soviets did dominate Poland and had military divisions stationed there.

CARTER CONSULTS. In the 1980 campaign, it was Carter who fumbled a Cold War question during a presidential debate when he cited his 13-year-old daughter, Amy, on the subject of nuclear war. "I had a discussion with my daughter, Amy, the other day, before I came here, to ask her what the most important issue was," Carter said. "She said she thought nuclear weaponry ? and the control of nuclear arms. This is a formidable force." The debate audience snickered. Carter's rival, Republican Ronald Reagan, served up a perfect rejoinder at a campaign rally, telling the crowd: "I remember when Patty and Ron were little kids, we used to talk about nuclear power," Schroeder recounted. Carter allowed in his memoir that "It was obvious that I had not expressed myself well."

DUKAKIS' TWO-FER: Michael Dukakis' run against President George H.W. Bush in 1988 yielded two lulus. His emotionally detached answer to a debate question about whether he would favor the death penalty if his wife, Kitty, were raped and murdered was a classic case of being too cool under pressure. "I think you know that I've opposed the death penalty during all of my life," he calmly replied. "I don't see any evidence that it's a deterrent, and I think there are better and more effective ways to deal with violent crime." Kitty Dukakis later wrote in her memoir, "That chilling incident at the second debate was the nail in the coffin. ... Michael made a mistake; he answered a question he should have hurled right back into the face of his questioner."

And then there was that unfortunate photo of a helmeted Dukakis taking a spin in a tank ? the ultimate in What Not To Wear for candidates.

TIME OUT: The most telling moment in a three-way debate between Bush, Bill Clinton and Ross Perot in 1992, wasn't conveyed in words. It was Bush's glance at his watch. The president already was battling perceptions that he was out of touch and out of ideas in a time of economic distress. When the TV cameras caught him stealing a glance at his watch, it reinforced the impression that Bush wasn't up for the job. It didn't help, either, that when a young woman asked Bush how the national debt had affected him personally, he said he didn't really get the question.

MR. MISUNDERESTIMATED. George W. Bush served up enough malapropisms as candidate and president that it's hard to single out just one. But voters elected him twice, validating his theory that people "misunderestimated" him. That gave people eight years of mangled language and puzzling Bushisms to ponder. Among them: "I know the human being and fish can coexist peacefully." "Rarely is the question asked, is our children learning?" and "We look forward to hearing your vision, so we can more better do our job."

THE LITTLE PEOPLE: Condescension is one of the worst traps for a presidential candidate and Barack Obama stepped squarely into it in April 2008. Speaking to well-heeled donors at a private fundraiser in the liberal bastion of San Francisco, no less, Obama said voters in struggling small towns of Pennsylvania and the Midwest "get bitter, they cling to guns or religion or antipathy to people who aren't like them or anti-immigrant sentiment or anti-trade sentiment as a way to explain their frustrations."

When news of his comment leaked out, Obama got a boatload of grief from Hillary Rodham Clinton, his rival for the Democratic nomination, as well as presumptive Republican nominee John McCain. "Elitist and out of touch," snapped Clinton, whose supporters handed out "I'm not bitter" stickers. Much like Romney now, Obama defended the thought he was trying to convey before conceding he had chosen his words poorly.

It wasn't the first time the political sin of talking down to people had created problems for Obama's campaign. Earlier, he'd told Clinton during a debate, "You're likable enough, Hillary." And Michelle Obama set off her own tempest by declaring that the public's hunger for change powering her husband's presidential bid made her proud of her country "for the first time."

HTC's big 2011 was limited to those warm summer months, when everyone wanted a smartphone for the beach. Now the accountants have confirmed HTC's initial statement: fourth quarter revenues were down 2.49 percent (year on year). It made 101.42 billion Taiwanese dollars (just under $3.5 billion) in the last three months of the year, which looks worse than it is because of the blockbuster 135.8 Taiwanese dollar takings in Q3. Overall year on year revenue was up by 67.09 percent, but profits were down 11.88 percent, with the company saying the outlook won't get any better in the first quarter, but should pick up when it begins the process of shedding a few pounds phones.
